Studies in stroke patients or in laboratory animals have shown the critical role of the thalamo-cortical circuits in control of consciousness. Recently, however, the role of the thalamo-cortical circuits in cognition has been attracting much attention in studies to understand the neural basis of cognition. The mediodorsal thalamic nucleus (MD) is reciprocally connected with limbic circuitry including medial prefrontal cortex and amygdala, the key structures for fear formation and extinction. In addition to its distinctive anatomical relationship, the MD participates in learning and memory processing in fear extinction through thalamic dual firing mode: tonic and burst. On the other hand, ‘Eye movement desensitization and reprocessing’, a well-developed psychotherapy for trauma treatment, is based on dual stimulation: recalling traumatic memory while orienting to alternating bilateral stimulus (ABS). However, the neurobiological mechanism linking dual stimulation to fear memory modulation remains unknown. In my talk I will present evidence that the MD thalamic nucleus integrates dual-stimuli signals into fear extinction network in mice.
