▶Abctract
Fibroblast growth factor 21 (FGF21) is a hormone induced by various metabolic stresses that regulates energy homeostasis. In humans, SNPs in and around the FGF21 gene have been associated with macronutrient preference, including carbohydrate, fat and protein intake. Here we show that FGF21 administration markedly reduces sweet and alcohol preference. In mice, these effects require the FGF21 co-receptor -Klotho in the central nervous system (CNS) and correlate with marked reductions in dopamine and serotonin concentrations in the nucleus accumbens. Also, we recently have shown that FGF21 is induced by osmotic stress that cause dehydration and it plays an important role in thirst response. Altogether, our works suggest that FGF21 stimulates both water consumption and a behavioral drive towards drinking pure water, and is therefore a principal water regulatory hormone.
