▶Abctract
The cJun NH2-terminal kinase (JNK) signaling pathway contributes to inflammation and plays a key role in the metabolic disease and cancer. Macrophages are implicated in this response. To test the role of JNK, we established mice with selective JNKdeficiency in macrophages. The protection of the HFD-fed macrophage JNK-deficient (ΦKO) mice against insulin resistance is associated with reduced tissue infiltration by macrophages and failure of polarization to the activated M1 state. We observe that ΦKO mice exhibits reduced the expression of inflammatory cytokines, chemokines, reduced hepatic infiltration by inflammatory cells, and suppressed the development of hepatitis and hepatocellular carcinoma. JNK in macrophages causes inflammation and promotes both metabolic disease and the development of hepatocellular carcinoma. Together targeting macrophages with a drug that inhibits JNK may provide therapeutic benefit for the treatment of inflammatory diseases.
