▶Abctract
A new “Hippo” signaling pathway has been implicated in regulation of cell proliferation apoptosis, and tumorigenesis in Drosophila and mammalian. Hippo kinase, Salvador and Warts kinase (mammalian homologs: MST1/2, WW45, LATS1/2 respectively) are key molecules of the Hippo-pathway. Hippo-Sav-Warts restricts cell proliferation and promoting apoptosis in differentiating epithelia by inhibiting Yorkie (mammalian homolog, YAP), a transcription co-activator. The Hippo pathway in mammals has been extensively studied with respect only to epithelial developments and to control of organ size. Here, we have generated mice with liver-specific ablation of key Hippo components (Mst1/Mst2 and WW45, Lats1/Lats2). We will discuss the possible roles of the Hippo components in the regulation of hepatic stem/progenitor cells. Additionally, we will discuss that SRF-IL6 as critical mediator of YAP induced stemness in mammary epithelial cells and breast cancer. Specifically, SRF-YAP induces many MaSC signature genes, among which IL6 is critical target for YAP induced stemness. High SRF-YAP/TAZ expressions are correlated with MaSC-like, basal like breast cancer (BLBC) with high IL6 expression.
