The positioning of the nucleosome by ATP‐dependent chromatin remodelers provides the fundamental chromatin environment for the regulation of diverse cellular processes acting on the underlying DNA. Recently, genome‐wide nucleosome mapping has revealed more detailed information on the chromatin‐remodeling factors. Using genome-wide approaches, we analyzed total RNA transcriptomes, mRNA transcriptomes, histone modifications, ChIP-seq and nucleosome positioning profiles in the absence of CHD family. In this talk, I will describe how these chromatin remodelers regulate nucleosome occupancy and how these activities are linked to aberrant transcription and genomic stability. Finally, I will discuss the recent progress of our laboratory about chromatin dynamics by ATP-dependent chromatin remodeler in yeast and mouse embryonic stem cells.
