Obesity and insulin resistance are major factors in the development of type 2 diabetes and metabolic syndrome. Development of insulin resistance is deeply associated with increased lipid availability and obesity. Interestingly, lipodystrophic states, the opposite appearance to obesity, which are characterized by reduced fat mass and defective lipid storage in adipose tissue, are strongly associated with ectopic fat deposition and an almost identical constellation of metabolic problems associated with obesity. Fsp27, a member of the CIDE protein family, is almost exclusively expressed in lipid droplets of adipocytes. Recently, we found that the mice with Fsp27 deficiency have typical phenotypes of lipodystrophy. In this talk, I would like to discuss the similarity and difference of insulin resistance between general obesity and lipodystrophy.
