                *Abctract
             CARP-1/CCAR1 is a cytoplasmic phospho-protein that is a regulator of cell growth and apoptosis signaling. Our goal is to develop novel, safer, and effective anti-cancer therapies on the basis of cell growth and survival inhibiting functions of CARP-1. CARP-1 interacts with key cell signaling transducers including the Anaphase Promoting Complex/Cyclosome subunit (APC)-2 and the nuclear factor kappa B regulator NEMO/IKKgamma. A cell-permeable, multifunctional peptide, termed TAT-N-A, that has APC-2 and NEMO-binding CARP-1 epitopes, inhibits growth of cancer cells. By conducting the high-throughput screening of chemical libraries, we next identified novel, small molecule inhibitors of CARP-1:APC-2 interaction. These compounds, called CARP-1 Functional Mimetics (CFMs), inhibit growth of a variety of cancer cells including the drug-resistant cancer cells without affecting growth of immortalized mammary epithelial cells. CFMs suppress cancer cell growth in part by stimulating CARP-1 levels and promoting apoptosis. CFMs impact biological properties of cancer cells by inhibiting their movement, invasion, and growth in soft-agar. A nanolipid formulation (NLF) of CFM-4, our lead compound, enhances its bio-availability. Oral administration of CFM-4 NLF suppresses growth of Non-Small Cell Lung (NSCLC) and Triple Negative Breast Cancer (TNBC) xenografts. In conclusion identification of CARP-1 and CFMs provide proof-of-concept to facilitate development of strategies to combat a variety of cancers including the drug-resistant cancers.
