*Abctract
The dynamic nature of chromatin is functionally important for the regulation of diverse DNA-dependent processes in the nucleus including transcription, replication, and DNA repair. Our goal is to understand how histone modifications and rearrangements specify the different gene expression patterns in response to external variables and how they alter critical epigenetic events related to cellular malignancies, notably cancer development. I will present our recent results supporting that histone modifications and proteolysis act as integral components of gene regulation in human cells. Our results emphasize a novel aspect of chromatin reconfiguration in proper gene expression and illustrate the power of combined biochemical and cellular approaches for mechanistic analyses.
