                 *Abctract
             The INO80 chromatin remodeling complex functions in stressed DNA replication in yeast and also is important to maintain the genome stability of normally growing human cells. However, the role of INO80 in normal DNA replication has remained unclear and the regulatory mechanisms, particularly, how INO80 is targeted to replication forks, are unknown. Recent studies from our laboratory show that the INO80 complex promotes replication fork progression during normal DNA replication in mammalian cells. The studies also identify a protein that regulates INO80 for DNA replication and reveal a novel mechanism that targets INO80 to replication forks. Importantly, our studies suggest that this regulatory mechanism is associated with tumorigenesis of certain cancer. In this talk, I will present the data that demonstrate the role and mechanisms of INO80 in normal replication fork progression and also will discuss the clinical significance of our results
