Telomeres have been implicated in cancer and aging. In mammalian cells, telomeres are extended by the telomerase reverse transcriptase.  In addition, the telomeres are bound by the core telomeric proteins TRF1, TRF2, RAP1, TIN2, TPP1, and POT1. These six telomeric proteins form large protein complexes and recruit different signaling molecules for telomere length control and the protection of telomeres from being recognized as DNA double-stranded breaks. How signaling pathways and cell cycle impact telomerase biogenesis and telomere-binding proteins remains largely unexplored. We have carried out proteomic analyses to illustrate how core telomeric proteins assemble on telomeres to recruit the telomerase and additional factors for telomere maintenance and protection. Our findings highlight a complex signaling network that governs telomere biology in mammals.
