*Abstract
Modern biophysical tools bring unprecedented precision and clarity to the table, enabling new types of biological inquiries. Single molecule fluorescence imaging is one such example but a majority of its applications has been to in vitro systems. My laboratory is trying to bridge single molecular measurements to systems level understanding of biology. In the first part, I will describe SiMPull (Single Molecule Pull-down), a technique to capture cellular protein complexes and analyze their composition, stoichiometry and activities at the single complex level. In the second part, I will describe our efforts to decipher antiviral signaling in a highly quantitative manner. Our single molecule FISH (Fluorescence In Situ Hybridization) analysis revealed the precise timing of gene expression of key antiviral signaling molecules and provided a molecular basis for heterogeneous expression of interferon. We hope to build a holistic narrative of cellular decision making through single cell analysis of protein interactions network and signaling pathways with the ultimate resolution of single molecules.
